چکیده
Abstract: Background: Recently, a lot of interest has been focused on the discovery high L-tyrosinaseyielding microorganisms that catalyze production enantiomerically pure L-DOPA, which recognized as conventional treatment for Parkinson's disease. Objectives: The current work primarily isolating novel, promising tyrosinase-producing isolates and performing taxonomical characterization them. Materials Methods: Initially six various actinomycetes were used in present study. Primary screening secondary selected performed. By evaluating morphological, cultural, physiological, biochemical molecular characterization, identified. Using 16S rRNA gene sequencing, isolates' Phylogenetic investigations conducted. Results: Streptomyces cellulosae variants, PD18 PD26 identified to be potential actinomycete their respective maximal tyrosinase yields reported 21.20 IU/mL 24.19 IU/mL. Conclusion: According observations experiment, SS (Soluble starch) medium with 1mg/mL copper sulphate was most suitable from our PD26. maximum yield noticed after 168 hr incubation. This emphasizes significance finding novel variants cellulosae, first produce well sources potentially bioactive substances like antimicrobial metabolites, biosurfactants enzymes amylase, lipase, cellulase, uricase, urease, β- galactosidase L- asparaginase. Keywords: Tyrosinase, tyrosine, medium, sulphate, cellulosae.

Streptomyces Tyrosinase: Production and Practical Applications
Tyrosinases (monophenol, o-diphenol:oxygen oxidoreductase, EC (1.14.18.1) are copper-containg enzymes which catalyze the o-hydroxylation of monophenols and subsequent oxidation of o-diphenols to quinones. The enzymes are involved in the pigmentation and are important factors in wound healing and primary immune response. Tyrosinases are found in prokaryotic and eukaryotic microorganisms, in mamm...

Background and Aims: Citrus exocortis disease is naturally limited to citrus plants and causes a serious disease on sensitive rootstocks such as trifoliate orange. The aim of this study was to sample new variants of citrus exocortis viroid from Fars trees with yellowing and suberization symptoms and comparison with other isolates of this viroid. Materials and Methods: A number of symptomat...

L-asparaginase is an anti-neoplastic agent used in the chemotherapy of lymphoblastic leukaemia. The present work deals with production of extra-cellular Lasparaginase from marine actinomycetes, using submerged fermentation. Marine actinomycete Streptomyces associated with marine spongeCallyspongia diffusa was isolated using specific ISP medium. Sponge-associated Streptomyces was character...
متن کاملProduction, purification and characterization of l-asparaginase from streptomyces gulbargensis
L-asparaginase is an anti-neoplastic agent used in the lymphoblastic leukaemia chemotherapy. In the present study a novel strain, Streptomyces gulbargensis was explored for the production of extra-cellular L-asparaginase using groundnut cake extract. The optimum pH, temperature, inoculum size and agitation speed for enzyme production were pH 8.5, 40°C, 1x10(8)spores/ml and 200 rev/min respectiv...
متن کاملAlbino mutants of Streptomyces glaucescens tyrosinase.
Site-directed mutagenesis was used to determine the functional role of several residues of Streptomyces glaucescens tyrosinase. Replacement of His-37, -53, -193 or -215 by glutamine yields albino phenotypes, as determined by expression on melanin-indicator plates. The purified mutant proteins display no detectable oxy-enzyme and increased Cu lability at the binuclear active site. The carbonyl d...
